芦荟大黄素抑制人胃癌细胞株SGC-7901增殖研究 被引量:5

摘要 目的:研究芦荟大黄素对人胃癌细胞株SGC-7901的增殖抑制机制。方法:采用MTT方法观察芦荟大黄素对人胃癌细胞株SGC-7901的增殖抑制作用;采用流式细胞仪技术检测细胞周期和DNA含量;应用免疫组化方法观察PCNA的表达。结果:芦荟大黄素能抑制人胃癌细胞株SGC-7901的增殖(P<0.05);使G_0/G_1期细胞增多,使S期和G_2/M期的细胞明显减少,细胞增殖抑制率明显增高,DNA指数明显降低(P<0.05);芦荟大黄素能抑制PCNA的表达(P<0.05)。结论:芦荟大黄素抑制人胃癌细胞株SGC-7901的增殖,可能是通过抑制PCNA的表达而影响细胞周期的结果。 Objective: To observe the inhibitory mechanism of aloeemodin on human gastric cancer cell Line SGC-7901. Method: The effects of aloeemodin on the proliferation of gastric cancer cell line SGC-7901 was evaluated by MTT; FCM was used to analyze the change of cell cycle arid DNA content ; Immunohistochemistry was used to examine expression of PCNA gene. Result: aloeemodin clearly inhibited the multiplication of SGC-7901 cells (P 〈 0.05 ) ;aloeemodin exclusively arrested cells at the G0/G1 phase, cells at the S and G2/M phase was significant decrease, the proliferation inhibition ratioyas higher than DMSO group, DNA index depressed significantly (P 〈 0.05 ) ; PCNA expression was reduced in aloeemodin groups, there was significant difference (P 〈 0.05 ). Conclusion: aloeemodin has significant inhibitory effect on SGC-7901. Aloeemodin inhibiting proliferation of SGC-7901 may be related to the reduced expression of PCNA which influence the cell cycle.
