摘要 [目的]探索海洋产几丁质酶微生物最佳的发酵组合。[方法]从唐山港曹妃甸港区海泥中分离筛选出2株几丁质酶活性强的菌株,研究不同的培养条件、诱变及组合发酵对菌株产酶的影响。[结果]通过紫外诱变产生的菌株的透明圈较大,直径在3~4mm,但数量不多;培养基碳源以胶体几丁质最好,产几丁质酶的最适温度在32℃;pH在7.0~8.0产酶量最高;以蛋白胨和酵母粉为氮源对产酶的效果明显;添加Mn^2+的培养基产酶能力最强;随着培养时间的增加,诱变的菌株组合发酵产酶后,几丁质酶活性显著提高。[结论]在组合发酵中,选取高产酶培养条件、不同来源的菌株混合培养对组合发酵有着重大的意义。 [ Objective ] The aim was to explore the optimum fermentative combination of marine producing chitinase microbes. [ Method ] 2 strains with strong chitinase activity were isolated from the sea mud from Caofeidian port area of Tangshan harbor and the effect of the different culture condition, induction and combined fermentation on producing chitinase of strain were studied. [ Result] The transparent circle of the strain produced by UV mutation was bigger and the diameter was 3 -4 mm, but the quantity was less. The colloidal chitin was the best carbon source of medium and the optimum temperature for producing ehitinase was 32 ℃ ; The yield of enzyme was highest when the pH value was between 7.0 and 8.0 ; the effect of using peptone and yeast as nitrogen source on producing chitinase was obvious ; the producing enzyme capability in the culture medium adding Mn^2+ was strongest; after the induced strain produced enzyme through combined fermentation, the chitinase activity was increased significantly with the increment of the culture time. [ Conclusion ] In combined fermentation, Selecting the culture condition of the enzyme with high producing ability and mixed culture with different source strain had great meaning for combined fermentation.
