PTEN和BAG-1蛋白在子宫内膜样腺癌组织中的表达及其临床意义

摘要 目的:通过分析PTEN、BAG-1在不同子宫内膜组织中的表达及与各临床病理因素之间的关系,探讨子宫内膜样腺癌发生、发展的分子生物学机制,为子宫内膜样腺癌的生物学特性的评定及靶向基因治疗寻找新的分子生物学指标。方法:采用免疫组化S-P法对60例子宫内膜样腺癌(E组)、20例非典型增生性子宫内膜(A组)和20例正常子宫内膜(N组)组织中两种蛋白行检测并分析。结果:PTEN的表达率:N组高于A组及E组,G1级高于G2+G3级,<1/2肌层浸润高于≥1/2肌层浸润(P<0.05)。BAG-1的表达率:N组低于A组及E组,G1级低于G2+G3级,无淋巴结转移低于有淋巴结转移(P<0.05)。PTEN与BAG-1在子宫内膜样腺癌组织中的表达呈负相关(r=-0.512,P<0.01)。结论:PTEN表达降低或BAG-1的高表达能促进子宫内膜样腺癌的发生、发展。PTEN与BAG-1两蛋白可通过负性调节对方的表达而间接发挥功能。 To investigate the expression of PTEN and BAG-1 in the different endometrium tissue and analyze the difference of the protein's expression and the relation between the clinicopathologie characters with expression in order to explore the machinery of molecular biology for endometrioid adenocarcinoma. The experience will look for the new markers for evaluating biological characters and gene therapy. Methods:We examined the expression of PTEN, BAG-1 among the 100 cases of endometrioid tissue. The results were treated by Wilcoxon test, Kruskal-Wallis test and Spearman correlation analysis. Results:The positive rate of PTEN was higher in normal group ( N ) than alypical hyperplasia endometrium ( A ) and endometrioid adenoearcinoma group ( E ) ( P 〈 0. 05 ).There was a relationship between the expression of PTEN with the histological grade and muscle invasive depth( P 〈 0. 05 ). The positive rate of BAG-1 was lower in N than that in A and E ( P 〈 0. 05 ). There was a relationship between the expression of BAG-1 with the histological grade and lymph node metastasis ( P 〈 0.05 ). The expression of PTEN negatively correlated with the expression of BAG-1 ( r = - 0. 512, P 〈 0, 01 ). Conclusion : The expression loss of PTEN or the increasing expression of BAG-1 could accelerate the generation and development of the endometrioid adenocarcinoma. PTEN make the function of restraining cancerization by regulating nagetively the expression of BAG-1 besides by its own action.
