纳米α-亚麻酸对病毒性心肌炎小鼠心肌损伤的保护作用 被引量:4

Protective effect of nano-α-linolenic acid in viral myocarditic mice

摘要 目的:探讨果王素活性成分纳米α-亚麻酸对病毒性心肌炎(VMC)小鼠心肌损伤的保护作用。方法:30只BALB/C小鼠腹腔无菌注射柯萨奇病毒(CVB3)制作病毒性心肌炎模型,造模后小鼠随机分为生理氯化钠溶液(NS)对照组和纳米α-亚麻酸高、低剂量治疗组(180,60 mg.kg-1),灌胃7 d,每组10只。另取20只BALB/C小鼠腹腔无菌注射病毒培养液,分别给予NS和纳米α-亚麻酸对照组(180 mg.kg-1,灌胃给药,7 d)。测定小鼠给药前后的心率变化率,计算死亡率;14 d后处死小鼠,并取血检测血清肌酸激酶同工酶(CK-MB)、心肌肌钙蛋白(cTn-T)和B-型钠尿肽(BNP)水平。结果:心肌炎小鼠NS对照组的心率增加率、血清CK-MB、cTn-T和BNP表达水平均显著高于非感染小鼠(P<0.01);与心肌炎小鼠NS对照组比较,纳米α-亚麻酸180 mg.kg-1治疗组小鼠心率增加率[(38.2±8.7)%vs(12.6±1.9)%,P<0.05]和死亡率均明显降低[50%vs10%,P<0.05],CK-MB,BNP和cTn-T表达水平均显著下降(P<0.01)。结论:纳米α-亚麻酸可以提高病毒性心肌炎小鼠的生存率,减轻心肌损害。 Objective: To determine the protective effect of nano-α-linolenic acid in viral myocarditic mice. Methods: Thirty BALB/C mice were infected with coxsackie virus B (CVB3 ) to develop viral myocarditis. Then the mice were treated with saline (control) , or 60 and 180 mg.kg^-1 nano-α-linolenic acid, ig, for 1 week, respectively. In another series, 20 BALB/C mice were treated with saline (control) and nano-α-linolenic acid 80 mg.kg^-1, ig, for 1 week(n = 10 for each group). The death rate was calculated; the serum levels of creatine kinase isoenzyme MB(CK-MB), cTn-T and B-type natriuretic peptide (BNP) were measured after last doses. Re- sults : The heart rate, and the serum levels of CK-MB, cTn-T and BNP increased in myocarditic mice. Nano-α-linolenic acid reduced the increased heart rate [ (38.2 ± 8.7)% vs (12.6 ± 1.9)% , P 〈0.05], the death rate [50% vs 10%, P 〈0.05 ], and the levels of CK-MB, BNP and cTn-T(P 〈0.01) in myocarditic mice. Conclusion: Nano-α-linolenic acid can increase survival and attenuate myocardial injury in viral myocarditic mice.
