摘要 目的:构建抗人胃癌全套单链抗体基因噬菌体表面呈现文库.方法:利用噬菌体表面呈现技术,构建基因文库,经过panning筛选富集后,用ELISA方法检测抗原结合活性.结果:从94个单个噬菌体克隆中一次筛选到18个具有抗原结合活性的阳性克隆.结论:我们所应用的方法为抗肿瘤基因工程抗体的制备提供了一条更为有效的新途径。 Aim: To construct the phage display library of the immunoglobulin repertoire of antihuman gastric cancer single chain Fv antibody. Methods:The ScFv gene library was constructed by phage display method. The affinity to antigen was detected by ELISA after panning. Results:Eighteen antigenbinding clones were identified from 94 individual phagemid clones. Conclusion: In the production of engineering antibodies against human cancer, this strategy may provide an alternative approach, and it also provides a basis for further studies on diagnosis and therapy of gastric cancer.
