成纤维细胞生长因子及血管内皮生长因子在喉粘膜上皮中的分布及作用 被引量:11

摘要 目的:探讨成纤维细胞生长因子超家族成员(bFGF和KGF)及血管内皮生长因子(VEGF)与喉粘膜上皮与间质相互作用的关系.方法:用免疫组化方法和原位杂交方法,分别观察bFGF,KGF及VEGF在喉部正常粘膜(N)和炎症(IF)中的分布.结果:bFGF和KGF的转录仅在N及IF的间质中出现,阳性反应位于血管内皮细胞和成纤维细胞,此外巨噬细胞也见阳性反应,上皮细胞均未见阳性表达.而VEGF不仅在上皮细胞中表达,部分血管内皮细胞及慢性炎性细胞亦表达VEGF.在IF中,bFGF,KGF及VEGF的阳性率和反应程度较N均有所提高.结论:bFGF,KGF和VEGF通过自分泌或(和)旁分泌途径促进上皮细胞生长及间质中的血管形成,在上皮与间充质细胞的交互作用中发挥着重要的作用. Aim: To determine the relationship between the members of fibroblast growth factor (FGFs),VEGF and the epithelialmesenchymal interaction of laryngeal mucosa Methods: The distributions of bFGF, KGF and VEGF in laryngeal normal mucosa (N) and chronic inflammatory mucosa (IF) were observed by in situ hybridization and immunohistochemistry Results: The mRNA transcriptions of bFGF and KGF were present in the mesenchyme, but not in epithelial cells The positive staining of bFGF and KGF were detected in fibroblasts, endothelial cells and macrophages Whereas, the expression of VEGF was located not only in epithelial cells, but also in vascular endothelial cells and some chronic inflammatory cells The expressions of bFGF, KGF and VEGF showed increased level in IF than in N Conclusion: bFGF, KGF and VEGF could not only enhance neovascularisation, formation of mesenchyme and proliferation of epithelial cells in autocrine or/and paracrine way, but also function in the local inflammatory and immunologic reaction BFGF,KGF and VEGF play important roles in the epithelialmesenchymal interaction
