摘要
应用薄层荧光扫描法测定了动物血浆中佐匹克隆的浓度,并对佐匹克隆在中毒家兔体内的分布进行了研究。样品在pH686条件下用二氯甲烷—异戊醇(98∶2)提取,氯仿—甲醇(9∶1)展开,用CS—930薄层扫描仪在激发波长305nm进行荧光单波长线性扫描测定。本法对血浆中佐匹克隆的最低检出限为25ng/mL,平均提取回收率80%,RSD为204%。研究发现,急性中毒家兔12h后其胃内容物、肝、脑和胆汁中佐匹克隆含量较高。
A method is reported for the detection and analysis of zopiclone concentration in plasma by thin-layer chromatography-fluorometry and used to study the distribution of zopiclone in poisoned rabbits.The samples were adjusted to pH 686,extracted with dichloromethane-isoamyl alcohol(98∶2)and developed with chloroform-methanol(9∶1).The plate was scanned at an excitation wavelength of 305nm and number two emission filter plate in the fluoresence linear scan mode of CS-930.The average recovery was 80% and RSD was 204%.Its limit of detection in plasma was 25ng/mL.The concentrations of zopiclone in gastric contents,liver,brain and bile were higher than these in other tissues when the rabbits were poisoned after 12 hours.
