流式细胞仪在异体外周血干细胞移植中的应用

Using of flowcytometry in allo-peripheral blood stem cell transplantation

目的：研究外周血干细胞移植时，流式细胞仪在干细胞含量检测方面的应用。方法：用CD34单抗标记动员后CS－3000plus分离的外周血单个核细胞。用Facscalibur中的Cellquest软件收集分析。结果：正常人外周血中CD34+细胞从动员前的0．02％～0．22％上升到1．21％～2．47％。受者输入的CD34+量为3．71～11．8×106个细胞／千克受者体重。结论：流式细胞仪为临床提供了一种快速简捷的干细胞测定方法。

Objective: Research the use of flowcytometry(FCM) in allo-peripheral blood stem cell transplantation(PBSCT).Methods:donors' peripheral mononuclear cells which were marked CD34 antibody after moblilizationg were acquired andanalysed with cellquest version 1. 2. 2 software in Facscalibur. Results: concentration of Co34+ cells in healthy donors peripheral flood increased from normal level 0.02%～0.22% to 1 .21% ～ 2.47% and that of infusion CD34+ cells are3.71-11 .8 × 106 cells/kg recipient weight respectively.Conclusion: FCM is a rapid convenient method for detectingCD34+ Cells in allo-PBSCT.
