感染人体的博尔纳病病毒ORFⅠ基因前部片段的检测

Detection of the initial part of fragment of Borna disease virus ORF Ⅰ from human beings

目的感染人体的博尔纳病病毒（ＢＤＶ）第一开放阅读框架（ＯＲＦⅠ）基因前部片段的检测至今尚未见报道。通过对先期实验已证实存在ＢＤＶ感染的人体标本和来源于感染动物的病毒标准品进行检测，拟了解感染人和动物的该病毒片段是否存在差异。方法用套式逆转录／多聚酶链反应（ｎｅｓｔｅｄＲＴ－ＰＣＲ）同时检测１２例ＢＤＶ感染的精神病人和健康献血者的周围血单核细胞和１００ｐｇ／μｌ的ＢＤＶ－ＭＤＣＫ标准病毒株中ＢＤＶＯＲＦⅠ（ｐ４０）基因前部片段。结果１２例人体标本的检测全部呈阴性，而病毒标准品呈阳性结果。结论该研究结果提示，感染人体的ＢＤＶ与来源于动物的ＢＤＶ相比较可能在病毒ＯＲＦⅠ基因片段的前面部分存在着较大的变异或前部片段缺失。该现象值得今后进一步研究。

Objective The detection of initial part of ORF Ⅰ fragment of Borna disease virus (BDV) so far is not reported. The research is for understanding whether or not there is any difference in the fragment between from animal and human beings infected with BDV proved by previous research. Methods The initial part of ORF Ⅰ fragment of BDV from BDV MDCK and from peripheral blood cells (PBMC) from 12 infected psychiatric patients and volunteers were checked by nested RT PCR.[WT5”HZ〗 Results The results from the detection of human samples are all negative but it is positive from BDV MDCK. Conclusion It may suggest that there is some great divergence or even deletion of ORF Ⅰ initial fragment of BDV from human beings compared with that from animals.This result may be worthful for further research in the future.
