摘要
目的一组抗gp130分子的单克隆抗体已被制备。对这组单抗的生物学及功能特性的研究首次表明,gp130分子上不同位点涉及不同的gp130细胞因子的生物学活性。在本研究中,其中一个抗gp130单抗,B-K5,将被作进一步研究。方法采用gp130细胞因子OM和LIF,B-K5单抗以及对gp130细胞因子高度敏感的细胞株(XG4-CNTF),通过3H-TdR的掺入作生物学检测,以期进一步确定B-K5单抗的生物学特性。结果当OM和LIF的浓度各自为300pg/ml和100pg/ml时,B-K5单抗可抑制OM和LIF的生物学活性;但当OM的浓度上升至3ng/ml时,B-K5仍可抑制OM的生物学活性。相反,当LIF浓度在1ng/ml时,B-K5却不能抑制了,即使B-K5单抗的浓度已被提高。结论B-K5单抗主要影响OM的生物学活性。
Objective A group of anti gp130 monoclonal antibodies (McAbs) was prepared.The biological and functional characterization of these McAbs has found for the first time that the different epitopes on gp130 molecule were involved in different biological activities of gp130 cytokines.One anti gp130 McAb,B K5,will further be characterized in this study.\ Methods\ [WT5”BZ〗The gp130 cytokines OM and LIF,B K5 McAb. and a cell line(XG4 CNTF)highly sensitive to gp130 cytokines were used for biological assay by virtue of 3 H TdR incorporation in order to further determine biological characteristics of B K5 McAb.\ [WT5”HZ〗Results\ B K5 McAb inhibited the biological activities of OM and LIF when OM and LIF concentrations were 300pg/ml and 100pg/ml, respectively.But,B K5 could still inhibit the biological activity of OM when concentration of OM was increased to 3ng/ml,whereas B K5 could not while LIF was at the concentration of 1ng/ml even if concentration of B K5 was enhanced. \ [WT5”HZ〗Conclusion\ B K5 McAb affected mainly the biological activity of OM.
