摘要
用体外原代培养的人脐静脉内皮细胞(HUVEC)观察rTNFα、rIL-1对HUVEC产生IL-1的诱导效应及重组IL-1受体拮抗剂(rIL-1ra)的抑制效应。结果表明,rTNFα以剂量依赖的方式促进HUVEC释放IL-1;刺激8hIL-1的释放活性显著提高,24h达峰值。rIL-1也可促进HUVEC释放IL-1,随rIL-1浓度的增加,IL-1的释放活性也增加,刺激4h,IL-1的释放活性即可明显提高(P<0.01)。rIL-1ra可抑制由rIL-1引起的IL-1释放,提高rIL-1ra的浓度,上清液中IL-1活性相应减低,当rIL-1ra浓度达10mg/L时。
The studies were made on the effect of production of IL 1 in HUVEC induced by rTNFα,rIL 1 and the inhibitory effect of rIL 1ra.The results showed that rTNFα promoted HUVEC to release IL 1 in a dose dependent manner.When HUVEC was stimulated,the secretion of IL 1 rose significantly 8 hours later and reached peak 24 hours later.Recombined IL 1(rIL 1)promoted HUVEC to produce IL 1.The production of IL 1 rose in proportion to rIL 1 dose dependently.The activity of release of IL 1 rose obviously 4 hours after HUVEC was stimulated( P <0.01).rIL 1ra inhibited IL 1 secretion of HUVEC induced by rIL 1.Activity of IL 1 in supernatant decreased when the concentration of rIL 1ra increased at the same time.When rIL 1ra reached 10mg/L.activity of IL 1 could be inhibited almost completely.
