胆总管结扎引起胆汁淤积对小鼠肝脏细胞增殖的影响 被引量:2

摘要 目的:探讨胆总管结扎引起的胆汁淤积对小鼠肝脏增殖的影响.方法:采用胆总管结扎手术制备胆汁淤积小鼠模型;利用全自动生化分析仪检测小鼠血清肝脏生化指标水平;用免疫组织化学方法检测小鼠肝脏中增殖相关的指标的表达情况.结果:通过胆总管结扎手术成功制备了梗阻性胆汁淤积小鼠模型;小鼠血清生化指标以及病理组织学变化表明小鼠肝脏功能受损,胆总管结扎手术成功;胆总管结扎引起胆汁淤积后,肝脏细胞有丝分裂相增加,肝细胞和胆管细胞中增殖相关的标志物表达升高,表明细胞增殖活性增强.结论:胆总管结扎引起的胆汁淤积可以引起肝脏细胞以及胆管上皮细胞的增殖. AIM: To investigate the effect of biliary cholestasis on liver induced by bile duct ligation in mice. METHODS : Mice were subjected to bile duct ligation to establish cholestasis model. After biliary intervention, hepatic biochemical indicators in serum were detected with automatic biochemistry analyzer, hepatic pathological changes were observed with HE staining and cell proliferation related markers in liver were detected with immunohistochemistry. RESULTS: An obstructive cholestasis mouse model was successfully established. Serum biochemical and pathological changes indicated impaired liver function in bile duct-ligated mice. After cholestasis caused by bile duct ligation, mitotic phase in hepatocytes and the expression of proliferation markers in hepa- tocytes and biliary epithelial cells increased,indicating that proliferative activity of liver ceils increased significantly after obstructive cholestasis. CONCLUSION: Cholestasis induced by bile duct ligation promotes the proliferation of liver cells.
