摘要
目的:探讨制备、鉴定基于sp17的伪病毒肿瘤疫苗,为肿瘤的生物治疗策略提供新的方法。方法:在特定的条件下将肿瘤抗原多肽和DNA制备成模拟病毒样结构的颗粒,并以电镜、DNA阻滞试验、DNaseⅠ保护试验和免疫印迹(Western blot)对该疫苗进行制备鉴定。结果:在一定浓度的氯化钠溶液中,电荷比为2的制备条件下,该疫苗能形成均匀的颗粒样结构,并可有效介导瘤苗所携带基因的表达。结论:该研究成功制备了基于sp17的伪病毒肿瘤疫苗,为新型的肿瘤疫苗的设计提供了新的思路。
Objective:To prepare and identify the pseudovirus tumor vaccine based on sp17.Methods:The preparing parameters were identified by assays of DNA gel retardation, DNase Ⅰ protection, and identified by Western blot. Results:The vaccine was condensed into homogeneous particles at charge ratio of 2 in the NaCl concentration,which could effectively mediate expression of the genes carried by vaccine particles. Conclusion:Our study provides a good foundation for the design of tumor vaccine.
