摘要
目的通过检测肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)的变化,探讨它们在深静脉血栓形成中的作用,为临床中深静脉血栓形成的预防、早期治疗提出新的靶点。方法将35只16周龄雄性Wistar大鼠随机分为正常对照组(C组,n=5)和模型组(DVT组),DVT组又分为模型D2、D6、D8、D12、D24组,每组6只。采用手术结扎右侧股静脉方法进行造模。对照组及DVT组分别于造模后2 h、6 h、8 h、12 h、24 h取血。用放射免疫分析法测定血清中TNF-α和IL-6的含量。结果DVT组各时间点IL-6和TNF-α血清值均高于正常对照组(P均<0.01);在DVT组中,随造模时间的延长,IL-6及TNF-α血清浓度不断升高,在血栓形成12 h时,二者血清浓度达到高峰。除D8组与D12组组间比较无显著性差异外,其他模型组组间比较有显著性差异(P均<0.01)。结论细胞因子IL-6,TNF-α水平随深静脉血栓的形成过程而发生变化,可作为观察血栓形成进展的指标。
Objective It is to explore the function of TNF - α and IL- 6 on process of deep vein thrombosis (DVT) through investigating their changes, so to propose new targets of prevention and early treatment for DVT in clinic. Methods 35 male Wistar rats aged 16 weeks were divided randomly into control group (group C, n = 5 ) and model group (DVT group). DVT group was divided into group Dz, D6, D8, D12, D24, every group had 6 rats. The femoral veins of the rats were ligated to establish model of DVT. Blood samples of control group and modd group at 2 h, 6 h, 8 h, 12 h and 24 h after model making were taken to determine the content of serum TNF - α and IL - 6 by radioimmunoassay. Results The level of serum IL - 6 and TNF - α in every time of the DVT model groups were higher than those of the control group ( P 〈 0.01). The level of serum IL - 6 and TNF - α in the model groups were gradually increased with the thrombosis, and achieved peak after 12h of the thrombosis. The DVT model groups comparisons showed that, there were statistical significances in every group comparisons ( P 〈 0.01 ), except group D8 with group D12 ( P 〈 0.05 ). Conclusion The levels of serum IL- 6 and TNF - α which change with the process of acute vein thrombegenesis could be the target of observing the process of thrombogenesis.
