摘要
用抗氧化剂及自由基清除剂小檗胺(中药提纯单体化合物)对STZ诱发的大鼠糖尿病性白内障进行腹腔注射的预防实验结果显示:1)在白内障出现早期小檗胺给药组晶状体空泡出现时间比未给药的糖尿病组推迟2周.2)小檗胺有对抗诱发动物模型晚期晶状体混浊出现的功能,以3.48mg/kg体重剂量的效果最好.3)SOD,CAT,GSH-Px酶活性的动态变化,未给药的糖尿病组第2周即开始出现,两个剂量小檗胺给药组均比糖尿病组延迟2周出现.这与裂隙灯观察结果相吻合,但形态学变化晚于酶活性变化.这证明早期使用抗氧化剂及自由基清除剂小檗胺对动物实验性糖尿病性白内障的发生、发展有明显的预防作用.
The preventive effect of berbamine on oxidative damage in rat diabetic cataract was investigated.Diabetic rats induced by streptozotocin(STZ) were intraperitoneally injected with two different doses of antioxidant berbamine,i.e.1.74 mg/kg B.wt.(1/31.51 LD50) and 3.48 mg/kg B.wt.(1/17.75 LD50).(1) At the early stage of diabetic cataract,the appearance of vacuoles in the lens was delayed for two weeks in diabetic rats injected with berbamine.(2) At the twelfth week,about 81.25% of diabetic rats showed in their lenses the opacity degree of Ⅳ and above,but only 40% and none of the diabetic rats injected with two doses of berbamine accordingly were found to reach such a stage of opacity in their lenses.(3) In comparison with normal control the activity changes of superoxide dismutase and catalase in the lenses of diabetic rats occurred at the second week,and were delayed for another two weeks by injection of berbamine.The activity changes of GSHPx were different from SOD and CAT.From week 2 to week 4 its activity in the lenses of diabetic rats was higher than that of normal control and the diabetic rats injected with two doses of berbamine.From these results,it was quite obvious that the antioxidant berbamine could prevent or delay the formation of diabetic cataract.
