摘要
目的探讨versican基因及蛋白在体外培养的毛乳头细胞(dermal papilla cells,DPC)中的表达变化,并研究其对DPC凝集性生长特性的影响。方法利用酶消化法分离培养DPC,并进行体外培养传代,观察不同代次DPC凝集性生长特性的变化。应用Western blot、RT-PCR检测DPC的β-catenin、versican的表达情况,对比研究其与DPC凝集性生长的关系。结果随着DPC由低代向高代分化,其凝集性生长特性逐渐减弱,versican表达水平逐渐降低。Wnt途径上调versican基因的表达,能够使高代DPC表达β-catenin、versican增强。结论versican基因和蛋白水平的表达与DPC凝集性生长呈正相关;versican基因是调控DPC凝集性生长的重要基因,versican的表达对毛囊(hair follicle,HF)的形成起着十分重要的作用,并在维护HF周期中扮演重要角色。
Objective To detect the changes of versican expression in the cultivation of dermal papilla cells,and to investigate the effect of versican on agglutinative growth of dermal papilla cells. Methods Dermal papilla cells were cultivated by enzyme digestion method,and the characters of agglutinative growth of dermal papilla cells at different passages were observed. The expres- sion of versican was detected by Western blot, RT-PCR. The relationship between the expression of versican and the agglutinative growth of dermal papilla cells were investigated. Results The agglutinative growth of dermal papilla cells were wear off and the expression of versican was decreased,as the dermal papilla cells were developed from low passage to high passage. High passage dermal papilla cells exhibited agglutinative growth when the versican expression was up-regulated by Wnt signaling. Conclusion The gene and protein expression of versiean positively correlate with the agglutinative growth of dermal papilla ceils, versican is identified as a target gene of regulating the agglutinative growth of dermal papilla cells. The expression of versican plays an important role in the formation of follicle and maintenance of hair cycle.
