摘要
采用单因素实验和正交实验对深层发酵桑黄获得最大菌丝体量的营养条件和培养条件进行了较系统全面的研究,并在20L发酵罐上进行了放大实验。结果表明,桑黄最适培养基组成为:玉米粉20g/L,葡萄糖10g/L,豆饼粉10g/L,玉米浆5g/L,KH2PO41.5g/L;适宜发酵条件为:25℃,160r/min,培养基起始pH5.0,接种量10%,装液量120mL/500mL,摇瓶培养10d。20L罐上发酵8d后,菌丝体量为14.58±1.21g/L。
Nutritional requirements and culture conditions of Phellinus linteus to obtain the higher production of mycelial biomass were well investigated by single factor and L9(3^4 ) orthogonal array. Results showed that the optimal medium composition could be set as follow:20g/L of corn flour, 10g/L of glucose, 10g/L of bean cake flour,5g/L of corn steep, 1.5g/L of KH2 PO4.The suitable culture conditions were obtained as follow:25℃, rotating speed 160r/min, initial pH5.0, inoculating amount 10%, 120mL medium in 500mL flask.The yield of mycelial biomass could reach to the level of 14.58 ±1.21g/L for 8d in 20L fermentor under the optimal conditions.
