摘要
研究了培养温度、培养时间、初始pH和摇床转速等培养条件对所选大肠杆菌(E.coli No TN1.5885)产L-天冬酰胺酶的影响。实验结果表明,该菌株进行摇瓶培养最为适宜的培养条件为:培养温度37℃,培养时间为16h,初始pH为7.0~7.5,摇床转速约为300r/min,其比酶活力达到约406U/g,相比较优化前提高了约20%。
The effects of incubating conditions including incubating temperature, incubating time, initial pH and rotate speed of shake bed on Ecofi (No TN1.5885)accumulating the anti-cancer drug were studied, as that was L-asparaginase. The results showed that the specific activity of L-asparaginase reached 406U/g under the optimum incubating conditions of incubating temperature 37℃,incubating time16h, initial pH between 7.0-7.5 and rotate speed of shake bed 300r/min.The yield of L-asparaginase increased by 20% compared with that of before optimization.
