野败型水稻的核质互作与基因差异表达

Interaction Between Nucleus and Cytoplasm and Gene Differential Expression in Rice CMS-WA Type

运用差异展示（ＤＤ－ＲＴ－ＰＣＲ）方法对野败型（ｗｉｌｄ ａｂｏｒｔｉｏｎ）细胞质雄性不育系珍汕９７Ａ及其相应的保持系珍汕９７Ｂ基因产物进行比较分析，发现这两个核基因组完全相同的品系间约有１０％的基因表达有差异．选取一个差异表达明显的ｃＤＮＡ成员Ｂ１３２进行Ｎｏｒｔｈ－ｅｒｎ杂交发现，该基因在保持系中正常表达，在不育系中表达受到明显抑制，而在杂种一代中又正常表达，在恢复系中表达水平也很低，这是一种典型的核质互作现象．Ｓｏｕｔｈｅｒｎ杂交中，Ｂ１３２在ＨｉｎｄⅢ和ＢａｍＨⅠ酶切长度片段上，各品系间未呈现多态性．

About 10% of the whole genes express differently between rice CMS-WA line Zhenshan 97A and its conservative line Zhenshan 97B detected by DD-RT-PCR analysis. Furthermore, gene B132, gives a typical expression pattern of interaction between nucleus and cytoplasm. Compared to its expression in line B, gene B132 is highly inhibited in line A, while it shows common expression as Line B in F1, and its expression in restorative line is al- so as low as that in line A. B132 doesn't show RFLP within different lines in Southern hybridization patterns.
