C/EBPβ在矽肺形成中表达的探讨 被引量:1

摘要 目的探讨C/EBPβ(CCAAT-enhancer binding protein-beta)在矽肺形成中的表达及其意义。方法运用免疫组化方法检测C/EBPβ在24只矽肺模型小鼠和16只正常小鼠肺组织中不同阶段的表达,对C/EBPβ阳性细胞进行分析。结果造模10d明显可见C/EBPβ的核移位,C/EBPβ在矽肺小鼠肺组织中表达较正常肺脏组织明显增强,差异有统计学意义,且造模1~30d的大鼠和30d后的大鼠中C/EBPβ表达的程度明显不同。结论C/EBPβ在矽肺大鼠肺脏组织内的多种炎性细胞中明显表达,且早期在一定时间范围内随着时间延长,其表达增加,能客观反映免疫或炎症早期进展程度,提示C/EBPβ在早期矽肺结节发生和进展中可能发挥重要作用。 Objective To investigate the expression and significance of CCAAT-enhancer-binding proteinβ (C/ EBPβ) in the development of pathophysiology of silicosis. Methods The experimental silicosis rat-model was established by intratracheal instillation of silica. Fourty sprague dawley rats were randomly divided into two groups: the silica exposure group of twenty-four rats and the control group of fourteen rats. The expression of C/EBPβ was determined by im- munohistoehemistry method . The C/EBPβ positive cell type was analyzed. Results C/EBPβ-positive cells were localized in epithelium and inflammatory cells in lung tissue. C/EBPβ nuclear translocation was observed in the 1-30 days silicotic tissue. There was a significant over-expression of C/EBPβ in lung tissue of silicotic rats as compared with that in the normal ones. The expression of C/EBPβ in the group 1-30 days was significantly higher than that in the group 31470 days after silica instillation(P〈0. 05). Conclusion The over-expression of C/EBPβ in inflammatory cells can reflect early the condition of immune response. Our results suggest that C/EBPβ may play an important role in the initial development of the silicotie nodule.
