摘要
目的了解福建省宁德市少数民族地区结核分枝杆菌的基因型特征。方法采用基于IS6110的PCR分型方法,对宁德市少数民族地区的73株结核分支杆菌进行检测,用Gel-Pro analyzer4.0软件和NTSYSpc2.10e软件对DNA指纹图谱进行聚类分析。结果根据IS6110-PCR指纹图谱,73株结核分支杆菌共分为两个主要类型,其中Ⅰ型48株占65.8%,Ⅱ型(北京基因型)25株占34.2%。汉、畲两族病例菌株指纹均为Ⅰ、Ⅱ型,但在两型中的分布差异有显著性(P<0.01),畲族以Ⅰ型居多,占78.9%。I、Ⅱ型菌株耐药率分别为39.6%(19/48)、28.0%(7/25)。结论宁德市少数民族地区结核分支杆菌北京基因型呈一般水平的流行,汉、畲两族之间结核病存在近期传播关系,但畲族人群病例以Ⅰ型(非北京基因型)流行为主,且耐药率较高。
Objective To understand molecular epidemiological features of Mycobacterium tuberculosis in Minority Regions in Ningde, Fujian Province. Methods The IS6110-based PCR fingerprinting method was used to analyze the 73 strains of MTB isolated of Minority Regions in Ningde.These fingerprinting patterns were Clustered by Gel-Pro analyzer 4.0 software and NTSYSpc 2.10e software,and analyzed logistic with epidemiological data from the patients. Results Based on the IS6110-PCR fingerprinting patterns of 73 strains of MTB isolated in Ningde, the genotypes of the isolated strains could be divided into 2 main types,in which the genotypes of 48 strains of MTB isolated was Ⅰ type, 25 strains was Ⅱ type, i.e. Beijing Genotype, and the genotypes in of Han and She nationalities were all types Ⅰ and Ⅱ, but the distribution of the genotypes between Han and She Nationality was significantly different (P〈0.01), the genotypes in the She Nationality was mainly type Ⅰ accounting for 78.9%. and the drug resistance rate of types Ⅰ and Ⅱ of strains of MTB isolates were 39.6% (19/48) and 28.0% (7/25). Conclusion M.tuberculosis strains of Beijing Genotype are prevalent at a common level in Ningde and there is may be a connection of tuberculosis in the nationalities of Han and She in s short period of time. But the genotypes in the She Nationality is mainlytype Ⅰ, the drug resistance rate of type Ⅰ of MTB isolate strains is higher,suggesting that type Ⅰ strains of MTB isolates is associated with high prevalence of tubereulosis in She Nationality in Ningde City.
