摘要
目的:用原位移植方法建立裸鼠人胃癌转移模型。方法:以SGC-7901人胃癌细胞株裸鼠皮下移植瘤组织块为材料,将其移植到裸鼠胃壁手术制备的粘膜小囊中。结果:该模型肿瘤生长潜伏期为3~4周,原位瘤沿胃壁呈浸润性生长,以后迅速增大,出现淋巴结及肝脏转移,并伴发幽门梗阻及癌性腹水等征象,荷瘤鼠生存期为16~24周(中位数生存期为18周)。本方法原位成瘤率及局部淋巴结转移率均为100%,肝脏转移率为66.7%(8/12)。结论:原位移植建立的裸鼠人胃癌转移模型较好地表现了人胃癌的生长过程及浸润转移特征。
Objectives: To estabilsh a metastatic model of human gastric cancer in nude mice. Methods: Nude mice model for human gastric cancer was established by orthotopic transplantation of tumor tissue pieces into a small artificial 'pouch' in stomach wall. The tumor tissue was obtained from subcutaneous xenografts of SGC 7901 human gastric cell line in nude mice. Results: Orthotopic tumors developed 3~4 weeks after operation and grew extensively. Subsequently, lymph nodes and liver metastasis developed. This model has yielded 100% take rate,100% local lymphatic invasiveness, and 66.7% (8/12) liver metastasis. Moreover, most of the animals developed pylorochesis and ascites. The survival time of tumorbearing nude mice was 16~24 weeks (median time 18 weeks). Conclusions: This metastatic model could represent the characteristics of invasiveness and metastasis of human gastric cancer.
