次声脑损伤后脑组织谷氨酸转运蛋白3的改变及意义 被引量:1

摘要 目的:探讨8 Hz 130 dB次声作用后脑组织中谷氨酸转运蛋白3(EAAT3)的表达改变及意义。方法:SD大鼠40只,随机分为对照组及次声作用1、7和14 d组各10只,次声作用组采用8 Hz 130 dB的次声按规定次数作用。采用Western blot检测EAAT3蛋白,RT-PCR行EAAT3 mRNA测定。结果:EAAT3在正常脑组织中表达较丰富。次声作用1 d后EAAT3蛋白和mRNA水平下调,7、14 d持续下调。结论:在次声脑损伤后,谷氨酸在细胞外堆积,可能与EAAT3下调导致谷氨酸重吸收障碍有关,因此上调EAAT3可能对次声脑损伤后脑组织有保护作用。 Objective:To investigate the expression of the excitatory amino acid transporter 3 (EAAT3) in brain after infrasonic damage. Methods: Forty SD rats were randomly divided into control and infrasonic damage groups with diverse damage levels. Rats were exposed to 8 Hz 120 dB infrasound as designed. Westernhlot was used to detect the changes of the protein and the mRNA was detected through RT-PCR. Results: The expression of EAAT3 showed an infrasound dose dependent down-regulation after infrasonic damage. Conclusion: The excellular glutamate accumulation may be caused by the compromised uptake of EAAT3 after infrasonic brain damages. Therefore the up-regulation of EAAT3 may be provide neuroprotection after infrasonic damage.
