摘要 目的血管内皮细胞不仅是机体循环系统和组织间的重要屏障,而且具有重要的代谢和内分泌功能。本研究主要观察海水对血管内皮功能基因表达的影响。方法12只Wistar大鼠随机均分为腹部开放伤合并海水浸泡组(实验组)和单纯腹部开放伤组(对照组),分别于海水浸泡或术后2h采集腹部开放伤大鼠小肠系膜血管组织,以实时定量PCR芯片法检测两组血管内皮通透与张力、血管生成、内皮激活与损伤等相关的84个功能基因的表达差异。差异表达大于2倍视为表达上调或下调。结果与单纯腹部开放伤相比,腹部开放伤合并海水浸泡大鼠小肠系膜血管内皮77.38%(65/84)的功能基因表达水平发生了明显改变,分别有61.90%(52/84)和15.48%(13/84)的基因表达水平上调和下调。其中,趋化因子Cxcl2表达水平升高了29.82倍,L-选择素表达水平下降了46.59倍。全部血管通透性与张力相关基因(12/12)和凝血激活相关基因(6/6)表达改变大于2倍。结论海水浸泡广泛引起腹部开放伤大鼠血管内皮重要功能基因表达改变,内皮功能受损。 Objective Sea water is a distinct injurious agent for trauma victim. The course of a trauma might develop differently when the injury is complicated by sea water immersion. Vascular endothelium is the protective layer situated between the blood circulation and tissues, and it possesses metabolic as well as endocrine functions. The purpose of the present study is to investigate the effects of sea water immersion on the expression of functional genes in vascular endothelial cells. Methods Rats were divided into two groups: simple open abdominal injury group and open abdominal injury + sea water immersion group. Two hours after the injury both with or without immersion, the vascular tissue of small intestinal mesentery was harvested, and then the expression levels of a total of 84 functional genes, which were involved in the functions of permeability, tonicity, angiogenesis, endothelial cell activation and injury, in vascular endothelial cells were detected and compared between the two groups by real time quantitative PCR array. The expressions were considered as upor down-regulated when more than 2-folds of expression difference existed. Results Compared with that of the rats in simple open abdominal injury group, the expression levels were markedly changed in 65 of a total of 84 functional genes (77.4%) in the vascular endothelial cells from small intestinal mesentery of the rats in open abdominal injury + sea water immersion group, including 52 genes (61.9%) up-regulated and 13 genes (15. 5%) down regulated. The expression level of chemokine (C-X-C motif) ligand 2 was elevated by 29. 8 folds, and that of L-selectin declined by 46. 6 folds, respectively. The expression levels of all the genes concerning permeability, vessel tone and thrombin activity changed more than 2 folds. Conclusion Sea water immersion may invoke abnormal gene expression in vascular endothelial cells of small intestinal mesentery in rat.
