摘要
对本研究室自建的小鼠前胃癌(MFC)细胞系、株和小鼠树突状细胞肉瘤(DCS)细胞系、株的移植瘤组织和培养细胞进行nm23/NDPK活性、免疫组化染色及RNA杂交的研究。-32PUTP生成法和乳酸脱氢酶一丙酮酸激酶偶联法均证明这些瘤组织或瘤细胞存在NDPK活性。MFC、株转移率高者NDPK活性低,反之则高;而DCS系、株瘤组织NDPK活性高、低与瘤株转移率高、低无明显规律性关系。MFC和DCS各株细胞的NDPK免疫组化染色呈弱阳性,nm23RNA狭缝杂交(地高辛标记探针)均呈阴性。结果提示nm23/NDPK对MFC可能具有转移抑制的功能。
The nm23/NDPK was studied for its activity, content and mRNA level in both mouse forestomach carcinoma cell line (MFC)and mouse dendritic cell sarcoma cell line (DCS). The NDPK activity was consistently identified in these tumor tissues or cells using assay of r32P UTP product and coupled pyruvate lactate dehydrogenase system. It was found that NDPK activity demonstrated an inverse relationship with the metastatic potential of the substrains from MFC, but variably related with that from the DCS. The cells of both MFC and DCS had weak positive staining immunohistochemically and negative in RNA slot hybridization using DIGlabelled probe. The results indicate that nm23/NDPK might play an important inhibitive role in the metastatic potential of MFC, but not in DCS.
