摘要
用荧光分光光度法测定马钱子碱(B)在小鼠体内的血药浓度及其药代动力学参数。结果表明:小鼠尾静脉注射3种剂量10,7.5和5mg·kg-1的B后时量曲线均符合二室开放模型。其主要动力学参数:分别为0.17,0.22和0.16h;分别为1.96,2.11和2.06h;Vc分别为7.23,7.59和5.65L·kg-1;AUC分别为2.48,1.96和1.45h·mg·L-1;CLs分别为4.03,3.82和3.44L·h-1·kg-1。此法为人体内的药代动力学研究提供了实验方法。
A spectrophotofluorometric method for determination of brucine concentration in blood and its pharmacokinetic parameter in mice was established. After brucine was iv administered at the doses of 10,7. 5and 5 mg·kg-1 to mice, the curve of blood concentration-time was shown in accordance with an open two-compartment model. The pharmacokinetic parameters of the three doses were as follows: were 0. 17,0.22 and 0. 16 h; were 1 .96,2. 11 and 2.06 h; Vc were 7.23,7.59 and 5.65 L·kg-1; AUC were 2.48,1.96 and 1. 45 h·mg· L-1; CLs were 4. 03, 3. 82 and 3. 44 L·h-1·kg-1, respectively. This method provides basis for clinical pharmocokinetics study.
