摘要
目的:研究2,3-吲哚醌(MW147)对动脉粥样硬化的抑制作用,并初步探讨其作用机制。方法:取雄性鹌鹑120只,随机分为正常对照组、模型组、阳性对照洛伐他汀(79.5mg·kg-1)组及MW147低、中、高剂量(20、60、120mg.kg-1)组。除正常对照组喂饲基础饲料外,其余组均喂饲高脂饲料,同时各用药组均灌胃给药连续8周,于8周末测定各组血清及组织中各脂质指标,包括胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白(L-DLC)及高密度脂蛋白(H-DLC)等;对主动脉及肝脏行光镜组织学检查,检测各组血清总超氧化物歧化酶(T-SOD)、谷胱甘肽过氧化物酶(GSH-Px)、总抗氧化能力(T-AOC)及脂质过氧化物丙二醛(MDA)水平。结果:与模型组比较,MW147可降低TC、TG、L-DLC、MDA水平,升高H-DLC、T-SOD、GSH-Px、T-AOC水平(P<0.05,P<0.01);主动脉及血脂中TC、TG水平也明显降低(P<0.05,P<0.01);病理结果显示主动脉内膜粥样硬化斑块的病变程度明显减轻。结论:MW147预防用药可抑制动脉粥样硬化形成,其作用机制与其降血脂和抗氧化作用有关。
OBJECTIVE: To study the possible mechanisms of the effects of indol - 2,3- dione (MW147) on experimental atherosclerosis (AS) . METHODS: A total of 120 male quails were randomly divided into six groups: normal control group, model group, lovastatin (79.5 mg·kg^-1) positive control group, and MW147 (20, 60, 120 mg·kg^-1) groups. The normal control group was fed on normal diet, while the other 5 groups were fed on high lipid diet and treated ig with corresponding drugs for eight weeks. Then the lipid levels including TC, TG, L-DLC and H-DLC in serum and tissues, and the total su- peroxidedismutase (T- SOD), glutathione peroxidase (GSH- Px), total antioxidant capacity (T- AOC) and malondialdehyde (MDA) were determined. Meanwhile the tissues of aorta and liver were observed under light microscope. RESULTS: In MW147 - treated groups compared with model group, the levels of TC, TG, LDL - C and MDA were decreased while the levels of HDL-C, T-SOD, GSH- Px and T- AOC in serum were increased (P〈0.05, P〈0.01); the levels of TC and TG in aorta walls and blood lipids were decreased obviously (P〈0.05, P〈0.01), and the pathological lesions of atherosclerotic plaque in MW147 groups were significantly lessened. CONCLUSION : The inhibitory effect of MW147 on atherogenesis might be related to its antioxidant and serum lipid- lowering action.
