二甲双胍对去势雌性大鼠血清性激素和瘦素含量的影响

Effects of Metformin on Serum Sex Hormone and Leptin in Ovariectormized Female Rats

目的:研究二甲双胍(MF)对去势雌性大鼠血清性激素和瘦素含量的影响。方法:取40只SD大鼠随机分为正常对照假手术组(NS5mL·kg-1·d-1,NS组)、单纯去势模型组(NS5mL·kg-1·d-1,去势组)、去势+MF低、高剂量组(MF135、270mg·kg-1·d-1,MF低、MF高组)。除NS组进行假手术外,其余各组手术去势,再连续灌胃给药30d,放射免疫分析法测定大鼠血清雌二醇、孕酮、黄体生成素(LH)、卵泡刺激素(FSH)和瘦素的含量。结果:与NS组比较,去势大鼠血清雌二醇、孕酮的含量显著降低(P<0.01),而LH、FSH和瘦素含量则显著升高(P<0.01)。与去势组比较,MF低、MF高组血清LH含量明显降低(P<0.05或P<0.01),MF高组瘦素含量明显降低(P<0.05),雌二醇、孕酮含量则显著升高(P<0.05);但MF对FSH含量无明显改变(P>0.05)。结论:MF可显著改善去势雌性大鼠性激素的分泌,降低血清瘦素的含量。

OBJECTIVE： To study the effects of metformin （MF） on serum sex hormone and leptin contents in ovariectormized female rats. METHODS： 40 SD rats were randomly divided into four groups： control group（normal saline 5 mL ·kg^-1·d^-1, NS group）, OVX group（normal saline 5 mL ·kg^-1·d^-1）, OVX ＋ low and high dosage MF group（135, 270 mg ·kg^-1·d^-1,MFlow and MFhigh group）,all group were ig drugs for 30 days on end.The serum sex hormone indexes including E2, P, LH, LSH and leptin content were measured with RIA. RESULTS： Compared with NS group, the serum E2 and P content were significantly lower（P 〈 0.01）, hut LH, FSH and leptin significantly higher in OVX group（P 〈 0.01） . Compared with OVX group, MFlow and MFhigh group decreased serum LH content （P〈0.05, P〈0.01）, MFhigh group also decreased serum leptin （P 〈 0.05） while increased serum E2, P content significantly（P 〈 0.05）; but MF had no effect on serum FSH content （P 〉 0.05） . CONCLUSIONS： MF can significantly improve sex hormone secretion and lower serum content of LEP in OVX rats.
