摘要
目的:建立以反相高效液相色谱法测定亚甲蓝微乳中主药含量的方法。方法:色谱柱为CenturySIL BDS C18,流动相为磷酸盐缓冲液(pH2.0)-乙腈-三乙胺(70∶30∶0.5),检测波长为292nm,流速为1.0mL·min-1,柱温为室温,进样量为20μL。结果:亚甲蓝检测浓度的线性范围为1.11~44.4mg·L-1(r=0.9992);平均回收率为101.2%,RSD=0.85%(n=6)。结论:本方法快速、简便、准确,可用于该制剂中主药的含量测定。
OBJECTIVE: To develop a RP - HPLC method for determination of main components in methylene blue microemulsions. METHODS: Sample separation was performed using a CenturySIL BDS C18 and monitored at 292 nm. The mobile phase consisted of pH 2.0 phosphate buffer- acetonitrile- triethylamine (70 : 30 : 0.5) pumped at a flow- rate of 1.0 mL·min^-1. The column temperature was kept at the room temperature and the sample size was 20uL. RESULTS: The linear range of methylene blue was from 1.11-44.4 mg·L^-1 (r = 0.999 2) . The average recovery was 101.2%, RSD = 0.85% (n = 6). CONCLUSIONS: The method is rapid, simple and accurate, and suitable for the determination of main components in this preparations.
