摘要
在热水抽提实验基础上,通过L9(43)正交实验,对从巴斯德毕赤酵母提取还原型谷胱甘肽(GSH)的实验进行优化,得到了最佳提取条件:1∶5的物水质量体积比,沸腾15min,加入1%的高氯酸后静置1.5h。实验结果表明:在热水沸腾15min后,溶液中GSH含量为67mg/100mL;加入1%高氯酸后,GSH平均浓度提高到152mg/100mL。采用截流相对分子量1k的超滤膜处理酵母提取液,降低了样品中大量杂质和大分子肽的含量,利于后续GSH的纯化。通过对离子交换树脂的吸附容量和解吸得率的研究,确定离子交换树脂D818型和HD-8型为GSH的分离介质。
The experiment for extracting reduced glutathione (GSH) from Pichia pastoris yeast based on hotwater extraction method was optimized through L9(4^3) orthogonal design. The optimal conditions were the ratio between fresh yeast and hot-water as 1:5, boiling for 15 mins, then added 1% perchloric acid and stayed for 1.5 hours at room temperature. The results indicated that the final yield of GSH obtained by the boiling process is 67 mg/100 mL, by adding 1% perchloric acid increased to 152 mg/100 mL. By ultrafiltration with cut off point of lku membrane for purifying the yeast extract, the content of the impurity and larger peptides in the sample decreased significantly, which is favor to subsequent purification of glutathione(GSH). By study of the adsorption capacity and desorption rate in ion-exchange resin chromatography, ion-resins D818 and HD-8 were selected as medium for GSH separation.
