氧化型低密度脂蛋白对胎盘生长因子及抑制剂的影响

摘要 目的探讨氧化型低密度脂蛋白(ox-LDL)对胎盘生长因子1(PLGF-1)作用人脐静脉内皮细胞(ECV-304)释放炎性因子和内皮细胞黏附分子的影响,及PLGF-1拮抗剂2’-氨基-3’-甲氧黄酮(PD98059)对其抑制程度。方法应用不同浓度的PLGF-1(20、40、80μg/L)孵育ECV-304,分为对照组和PLGF-1干预组(20、40、80 μg/L)。再应用PD98059(100μmol/L)和PLGF 1(80 μg/L)刺激ECV 304,分为空白对照组、ox-LDL组、PLGF-1+ox-LDL组和PLGF-1+ox-LDL+PD98059(拮抗组)。应用ELISA法检测炎性因子和内皮细胞黏附分子的表达。结果 (1)PLGF-1诱导炎性因子和内皮细胞黏附分子的表达呈时间依赖关系,6 h达最理想的分泌量。随着PLGF-1浓度的加大呈上升趋势。(2)与空白对照组比较,ox-LDL组、PLGF-1+ox-LDL组和拮抗组白细胞介素6、基质金属蛋白酶2、可溶性细胞间黏附分子1、可溶性血管细胞黏附分子1的表达显著增加(P<0.01),拮抗组较PLGF-1+ox-LDL组的表达显著减少(P<0.01)。结论 PLGF-1可促使内皮细胞产生大量炎性因子,参与诱导内皮活化;抑制PLGF-1的生物活性可控制炎性的反应。 Objective To study the effect of oxidized low density lipoprotein (ox-LDL) on the release of inflammatory cytokines and endothelial cell adhesion molecules from human umbilical vein endothelial cells (ECV-304) induced by placental growth factor-1(PLGF 1) and the inhibitory action of specific inhibitor 2 '-amino-3 '-methoxyflavone (PD98059)on this effect. Methods ECV-304(2×105) were incubated in the presence ofPLGF-1(20.40.80μg/L). According to the concentration of PLGF-1, the cells were divided into following subgroups: control group and PLGF-1 intervention groups(20,40 and 80μg/L groups). After pretreatment with PD98059 for 2 h,the cells were cultured with PLGF-I(80 〉g/L) and divided into following subgroups:the control group, PLGF-1 intervention group, ox-LDL intervention group, PLGF-1+ox-LDL intervention group, PLGF-1 + ox-LDL + PD98059 intervention group (antagonism group). Levels of inflammatory cytokines and endothelial cell adhesion molecules were assayed using ELISA quality kit. Results PLGF-1 induced the secretion of inflammatory eytokines and endothelial cell adhesion molecules in dose-dependent and time-dependent manner, with optimal secretion occurring at 6 h. PLGF-1 induced effect could be attenuated by PD98059. Conclusions PLGF-1 can induce the increase in expression of inflammatory eytokines which are responsible for inducing endothelial activation. Inhibition of PLGF can reduce inflammatory reaction.
