摘要
目的探讨哇巴因和血管紧张素Ⅱ(AngⅡ)对自发性高血压大鼠(SHR)动脉平滑肌细胞(ASMCs)分泌心钠素(ANF)的影响。方法组织块种植法培养4只SHR和4只Wistar-Kyoto(WKY)大鼠胸主ASMCs,分别用1×10^(-9)、1×10^(-8)、1×10^(-7)mol/L哇巴因、AngⅡ干预2种大鼠ASMCs,放射免疫法测定干预前及干预后3、6、12、24 h不同时间点ANF水平。结果干预前,WKY大鼠ANF水平高于SHR,但差异无统计学意义。哇巴因干预后,2种大鼠ANF水平显著升高,WKY大鼠6 h达到峰值(87.97±26.90)pg/10~6cells,SHR 12 h达到峰值(120.23±23.60)pg/10~6cells;随哇巴因浓度增高,2种大鼠ANF水平呈浓度依赖性升高。Ang Ⅱ干预后,WKY大鼠ANF水平6 h达到峰值(86.78±18.83)Pg/10~6cells,随后逐渐下降;SHR的ANF水平呈时间依赖性升高;随AngⅡ浓度升高,ANF升高的程度减弱。结论哇巴因和AngⅡ对局部ASMCs分泌ANF有显著影响。
Objective To investigate the effect of ouabain and Ang Ⅱ on ANF secreted by arterial smooth muscle eells(ASMCs) in SHR. Methods The thoracic ASMCs isolated from 4 SHRs and 4 Wistar-Kyoto(WKY)rats were cultured. ASMCs received ouabain or Ang Ⅱ 1 × 10^9 tool/L, 1 × 10^8 mol/L or 1 × 10^ 7 mol/L. ANF levels were measured with radioimmunoassay before and 3, 6, 12 and 24 h after treatment with ouabain or Ang Ⅱ. Results Before using ouabain or Ang Ⅱ, ANF level in WKY rats was higher than that in the SHR, but the difference had no statistical significance. After using ouabain, ANF level in ASMCs of rats was remarkably higher than before , ANF level in WKY rats reached the peak after 6 hours(87.97±26.90) μg/10^6cells, and that in SHR reached the peak after 12 hours(120.23±23.60) pg/10^6eells. With increase in the concentration of ouabain, ANF level of ASMCs in rats increased in a concentration-dependent manner. After using Ang Ⅱ , the level of ANF increased significantly,the ANF level in WKY rats reached the peak after 6 hours(86. 78±18.83) pg/10^6cells and that in SHR increased in a timedependent manner. With the increase in concentration of Ang Ⅱ , the increasing tendency of the level of ANF slowed down. Conclusions The secretion of ANF in partial ASMCs of SHR was affected remarkably by ouabain or Ang Ⅱ.
