摘要
采用μ-BondapakNH2TM色谱柱,以乙腈∶水∶甲醇(70∶25∶5,V/V)为流动相,利用示差折光检测器的高效液相色谱法对湖北毛坝大木、毛坝小木和建始3种生漆中漆多糖的单糖组成成分进行了直接分离分析,方法简便、快速、重现性好,回收率在98.8%~103.6%之间,相对标准偏差在5%以下。
This paper reports the separation and determination of monosaccharide composition in lacquer polysaccharide (LPS) by HPLC. The five monosaccharides were analyzed on μBondapak NH_2TM column (300mm×7.8mm i.d., 10μm) and refractive index detection. The mobile phase was CH_3CN H_2O CH_3OH(70∶25∶5, V/V) flowing at a rate of 1.6mL/min. These monosaccharides were identified by their retention times and quantitatively determined by their peak areas. LPS samples were hydrolysed with 2.0mol/L CF_3COOH. The linear correlation coefficients were all over 0.9953. The average recoveries of monosaccharides ranged 98.8% 103.6% and relative standard deviations were below 5%. The method is simple, rapid, precise, and has been used satisfactorilly for analysis of the monosaccharides hydrolyzed from LPS, which were isolated from sap of three kinds of Lac tree (Maoba Damu, Maoba Xiaomu and Jianshi) in Hubei province.
