应用基因芯片技术筛选肝细胞癌相关基因

摘要 目的:应用基因芯片技术筛选肝细胞癌相关基因。方法:利用CapitalBio公司基因芯片,对肝细胞癌组织和正常组织基因表达谱进行检测,并利用生物信息学方法对检测结果进行分析。结果:按差异显著性标准,从16000条基因中筛选出在肝细胞癌组织中共同差异表达基因326个,其中已知基因194个,新基因132个。在筛选出的已知基因中,表达上调113个,表达下调81个,包括原癌基因和抑癌基因、免疫相关基因、细胞信号和传递蛋白相关基因、蛋白翻译和合成相关基因、代谢相关基因、细胞周期蛋白相关基因、细胞骨架相关基因、细胞凋亡相关基因、DNA合成和修复相关基因等。结论:基因表达谱芯片技术可以筛选出肝细胞癌表达异常的相关基因群,并高效对基因功能进行研究。基因表达谱的分析有助于研究肝细胞癌发病机制。 Objective: To screen the hepatocellular carcinoma associated genes by eDNA microarray. Methods: The company of CapitalBio gene chips were used to analyze gene expression profiles in hepatocellular carcinoma and normal tissues. The results were analyzed by bioinformaties methods. Results: By applying this eDNA microarray, 326 differentially expressed genes in hepatocellular carcinoma were screened out among the 16000 target genes, comprising 194 known genes and 132 new gcne. Among the known genes,upregulated and downregulated genes were 113 and 81 respectively, including oncogenes and tumor suppression genes, immune related genes, cell signal transduction related genes, protein translation and synthesis related genes, metabolism related genes, cell cycle related genes, cell skeleton related genes, cell apoptosis related genes,DNA synthesis related genes, DNA damage and repair-related genes. Conclusion: The analysis of the gene expression profile of hepatocellular carcinoma using eDNA microarray can rapidly select associated abnormal genes, and it can help to rapidly explore the genes function. Further analysis of the obtained genes will help to understand the molecular mechanism of the hepatocellular carcinoma.
