脾虚小鼠小肠上皮细胞蛋白质组特征及白参的干预作用研究 被引量:7

Proteomics study of the small intestinal epithelia of splenic asthenia syndrome mice and the adjustive action of dry radix ginseng

摘要 [目的]研究脾虚型小鼠小肠上皮细胞蛋白质组特征及白参的调节作用。[方法]昆明种小鼠60只,利血平法复制动物模型,随机分为正常、脾虚模型和白参汤剂组,双向凝胶电泳技术分离蛋白质,改进的考染法染色,以PDQuest软件分析图谱,基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)进行质谱分析。[结果]鉴定出的33个蛋白质点中有27个表达明显减弱,6个表达增强。经白参汤剂治疗后,其表达多数得以回调。各蛋白质的功能类型几乎涉及小肠细胞各主要功能。[结论]脾虚型小鼠小肠上皮细胞的蛋白质组特征主要表现为蛋白质表达呈双向变化,以下调为主。表达变化的蛋白质涉及细胞各部位,白参汤剂对小肠上皮细胞蛋白质组的调节作用是白参健脾的重要机制之一。 [Objective]To approach the character of proteomics of small intestinal epithelia of the splenic asthenia syndrome mice and the adjustive action of dry Radix Ginseng. [Methods]Sixty Kunming mice were divided randomly into 3 groups: normal group, model group and dry Radix Ginseng decoction group. Splenic asthenia syndrome animal model were duplicated by reserpine. Two dimensional polyacrylamide gel electrophoresis technology was applied to separate proteins of small intestinal epithelia of mice of every group. The proteins spots were visualized with an improved technique of Coomassie brilliant blue staining. The pictures of proteins of every group were analyzed with the PDQuest software; the mass spectrometry analysis was used with MAL- DI-TOF-MS. [Results]The expression of 27 protein spots decreased and that of 6 proteins spots increased of 33 identified protein spots. After treated with traditional dry Radix Ginseng decoction, their increasing expressions of the proteins spots mostly were down-regulated and their decreasing expressions of the proteins spots mostly were up-regulated. The functions of identified proteins was almost related to the main functions of small intestinal cell. [Conclusion ]The chief character of proteomics of small intestinal epithelia of the splenic asthenia syndrome mice is bidirectional change, and down-regulation is dominant. The change of proteins are expressed in different parts of the cell. The adjustive action of dry Radix Ginseng of Chinese traditional medicine on proteomics of small intestinal epithelia is one of the important mechanism of invigorating spleen of dry Radix Ginseng.
