摘要
观察中药复方糖心康对糖尿病心肌损伤大鼠的作用机理。方法:采用链脲佐菌素(STZ)加高热量饮食建立糖尿病大鼠心肌损伤模型。采用美国Affymetrix Rat2302.0基因表达谱芯片及聚类分析,筛选差异表达基因,采用荧光定量RT-PCR法验证。结果:花生四烯酸12-脂加氧酶mRNA(Alox12 mRNA)是筛选出的差异表达基因。在糖尿病状态下,Alox12 mRNA基因表达上调,经糖心康治疗后表达下调。结论:Alox12 mRNA和糖尿病心肌损伤有重要相关性,糖心康对其有良性调节作用。
This is a study to observe the therapeutic effects of a complex TangXinKang prescription on DC rats. The model rats male Wistar were established with multiple injections at a low dose of STZ and high calorie diet. A range of examination methods were applied, including Affymetrix Co.US. Rat 2302.0 gene expression profiling chip, and cluster analysis, to observe the effects of Tangxinkang on the selected indicators. RT-PCR was also applied in the confirmatory experiment. It is found that Alox12 mRNA gene was a diabetic cardiomyopathy tissue with different expression. The Alox12 mRNA genes saw an up-regulation in the DC model group, and a down-regulation in the TangXinKang treatment group. It is concluded that the Alox12 mRNA was associated with Diabetic cardiac muscle damage, and that TangXinKang had a benign effect on Alox12 gene expression.
