摘要
目的观察年龄对大鼠大脑胆碱能神经系统中乙酰胆碱转移酶(choline acetyltransferase,ChAT)与乙酰胆碱酯酶(acetylcholinesterase,AChE)活性的影响。方法Sprague-Dawley雄性大鼠,按照年龄分为4~5周龄组、4~5月龄组和14~15月龄组共3组,每组10只。用比色法测定大鼠皮质、海马和纹状体区ChAT与AChE的活性。结果与4~5月龄组大鼠和4~5周龄组大鼠大脑皮质、海马和纹状体区ChAT活性比较,14~15月龄组大鼠明显降低(均P<0.01);4~5月龄组与4~5周龄组大鼠大脑皮质、海马、纹状体区ChAT活性比较,均没有显著性差异(P>0.05);与4~5月龄组和4~5周龄组比较,14~15月龄组大鼠3个脑区AChE活性明显降低(分别为P<0.05,P<0.01);4~5月龄组与4~5周龄组大鼠3个脑区AChE活性比较,均没有显著性差异(P>0.05)。结论年龄与胆碱能神经系统中ChAT和AChE的活性呈负相关。
Objective To investigate the effect of age on activity of cholineacetyltransferase (CHAT) and acetylcholinesterase (ACHE) in cerebral cholinergic nerve system of rat. Method According to age,male Sprague - Dawley rats were divided into the following 3 groups at random: 4 -5 weeks old group, 4 -5 months old group, 14 - 15 months old group (n = 10, respectively). Chromatometry was used to assay the activity of ChAT and AChE in cerebral cortex,hippocampus and corpus striatum. Results In 14 - 15 months old rats, activity of ChAT and AChE in cerebral cortex,hippocampus and corpus striatum was prominantly reduced, as compared with that in 4 - 5 months old rats and in 4 - 5 weeks old rats ( CHAT: P 〈 0. 01 ; ACHE: P 〈 0.05 and P 〈 0. 01 respectively) ; However, activity of ChAT and AChE was not remarkly different between 4 -5 weeks old rats and 4 -5 months old rats(ChAT and ACHE: P 〉0.05). Conclusion There is a negative correlation between age and activity of ChAT and AChE in cerebral cholinergic nerve system.
