摘要 目的研究Hartley豚鼠腰椎间盘自然老化及退变过程中脊索性髓核向纤维软骨髓核转化的规律及其血液生化指标与腰椎间盘退变的关系。方法Hartley雌性豚鼠40只,分别为1、1.5、2、2.5、3月龄,每组各8只。通过HE观察豚鼠椎间盘髓核退变情况,测定其血清中蛋白多糖水平。结果2月组椎间盘形态即有退变。血清蛋白多糖含量随年龄的增长而下降,各组间差异显著。结论随着年龄的增长,椎间盘退变的同时,出现血清蛋白多糖含量的降低。随着椎间盘的不断老化,终板的软骨细胞向髓核迁移,脊索性髓核向心性皱缩并最终完全被纤维软骨性髓核取代。在Hartley雌性豚鼠椎间盘退变的过程中,这一转化完成得更快、更早。 Objective To study the transition regularity of notochordal nucleus pulpusus to fibroeartilaginous nucleus pulposus in aging and degenerative Hartley guinea pig lumbar intervertebral disc, as well as its relationship with degeneration of intervertebral disc, ancl to detect its blood biochemical change and investgate its early pathogenesis. Methods 40 female Hartley guinea pigs aged 1 - 3 months were divided into postnatal 1 months group,postnatal 1.5 months group, postnatal 2 months group, postnatal 2.5 months group and postnatal 3 months group,with 8 animals in each group. The degeneration of the nucleus pulposus was observed by Hematoxylin -cosin stain, and proteoglyean in serum was measured by ELISA. Results Remarkable degeneration of the intervertebral disc was seen in guinea pigs of 2 months old. The level of proteoglyean was lower in animals of 3months old than in animals of 1 month old, and decreased with age. Conclusion When the intervertebral disc degenerated with age, the value of serum proteoglyean decreased. Aging guinea pig undergoes a chronological transition from notoehordal NP to fibrocartilaginous NP. All form fibrocartilage tissue found in the NP was formed by ehondrocytes that had originated and migrated from the cartilage endplate. The origin of ehondroeytes proceeded in a eentripetaldirection from the periphery toward the center of the NP. Female Hartley guinea pigs in the intervertebral discdegenera -tion process completed the conversion faster and earlier.
