鸡IGF1R基因多态性与生长和体组成性状的相关性研究 被引量:19

摘要 类胰岛素生长因子1型受体对细胞的生长、分化、增殖有重要调控作用,以东北农业大学肉鸡高、低脂双向选择品系第十世代肉仔鸡为研究对象,采用PCR-RFLP方法,对鸡IGF1R基因外显子4的C→G(919C→G)和外显子13的T→C(2761T→C)两个突变进行多态性检测。结果表明,919C→G对股骨重有极显著影响(P<0.01),对5周龄体重有显著影响(P<0.05),对龙骨长、胫骨长和胫骨重有一定影响(P<0.1)。2761T→C突变位点对跖骨围、胫爪比率有极显著影响(P<0.01),对胫爪重、股骨重有显著影响(P<0.05),对7周龄体重有一定影响(P<0.1)。群体遗传学分析表明,919C→G、2761T→C两个位点的等位基因频率在两系间分布差异极显著(P<0.01)。因此初步推断IGF1R基因可能是控制鸡5周龄体重、骨骼生长发育的主(效)基因或与主(效)基因紧密连锁。 Insulin-like Growth Factor 1 Receptor plays an important role in growth, differentiation and proliferation of apoptosis of cells. The 10th generation divergent lines of lean and fat broilers bred by Northeast Agricultural University were used in this study. Two mutations were detected using PCR-RFLP method, one C→G mutation (919C→G) in the exon4 and the other T→C mutation (2761T→C) in the exonl3 of chicken IGF1R gene. Association analyses indicated that 919C→G significantly related to the femur weight (P〈0.01), related to 5 weeks body weight significantly(P〈0.05), related to keel length, shank length and shank weight (P〈0.1). 2761T→C significantly related to metatarsus circle, metatarsus and ungual ratio (P〈0.01), and related to metatarsus and ungual weight, femur weight (P〈0.05), and related to 7 weeks body weight (P〈0.1). The population genetics analyses showed that the allele frequencies of 919C→G and 2761T→C had a significant difference between the lean and fat broilers(P〈0.01). So it is concluded that IGF1R gene is the major gene to affect 5 weeks body weight, growth and development of bone or linked to the major gene.
