钩体39kd疏水外膜蛋白疫苗对豚鼠神经体液和红细胞免疫功能影响的研究 被引量:2

摘要 为进一步为钩体基因工程疫苗的研究提供理论和实验依据,采用纯化的强毒017钩体39kd疏水外膜蛋白(OmpL39)疫苗免疫豚鼠,对其免疫力进行检定并观察该疫苗对神经体液和红细胞免疫功能的影响。结果显示:OmpL39在豚鼠体内能产生高效价的阳性抗体(MAT>1:3,200),免疫力合格;在免疫应答期,OmpL39疫苗对豚鼠外周血5羟色胺(5HT)、去甲肾上腺素(NE)、多巴胺(DA)等有明显影响,其中5HT、DA、NE明显降低,5羟吲哚乙酸明显升高,与空白对照组有显著性差异(P<005);OmpL39对红细胞免疫功能也有明显影响,其中红细胞cαb受体粘附酵母菌花环率,红细胞免疫复合物花环率均明显升高,红细胞免疫粘附因子促进率上升,抑制率下降,两者比值显著增大(P<005)。提示:在免疫应答期OmpL39除了增强体液免疫外,还能刺激机体影响神经递质的释放,对神经体液免疫功能有明显的调节作用。 A randomized control trial was conducted to determine the immunoprotective efficacy of OmpL 39 . 36 guinea pigs were divided into OmpL 39 group, whole leptospiral cell vaccine (WLCV) group, other proteins of Leptospira group, and negative control group (normal saline, NS). The results showed that all the guinea pigs of infected OmpL 39 and WLCV still survived, but all the control guinea pigs died. Immunoprotective efficacy was 100% for OmpL 39 and WLCV. OmpL 39 levels were similar to WLCV levels and higher than controls ( P <0 05). These suggested that OmpL 39 could be used as important immunoprotective antigen to develop the genetic vaccine of the targeting delivery system. Also it was observed that OmpL 39 could produce 100% immunoprotective efficacy, have higher MAT level(>1∶3200) and regulate 5 HT,5 HIAA, DA, NE. The 5 HT, DA, NE concentration was lower and 5 HIAA was higher than that of controls after stimulating the guinea pigs with OmpL 39 ( P <0.05). The results suggested that OmpL 39 genetic vaccine could produce immunity function and have an active effect on absorbing inflammation and protecting organs and tissues. Besides, the red cell immunity efficacy of the guinea pigs was changed during immunity response and the RBC C 3b RR and the RBC ICR were increased. The RFER was increased; the RFIR was decreased. RFER/RFIR was remarkably higer than that of control ( P <0.05). These suggested that OmpL 39 could increase red cell immunity function.
