益气活血中药对脑出血大鼠脑组织促血管生成素-1蛋白表达的影响 被引量:5

Effect of supplementing qi and activating blood circulation on expression of angiopoietin-1 in rats with intracerebral hemorrhage

摘要 目的:通过研究益气活血中药方剂对脑出血大鼠脑损伤区组织促血管生成素-1(Ang-1)表达的影响,探讨益气活血法治疗脑出血的作用机制。方法:155只SD大鼠被随机分为正常组5只,假手术组、模型组、益气活血组、益气组和活血组各30只。用Ⅶ型胶原酶诱导脑出血大鼠模型。各组相应灌服益气活血法代表方补阳还五汤全方及其益气组分、活血组分药物。各组于术后1、4、7、14、21和28d各取5只大鼠,采用Western blotting方法检测Ang-1的表达,结果进行光密度扫描半定量分析。结果:正常组及假手术组不同时间点Ang-1表达未见明显阳性信号;模型组、益气组、活血组及益气活血组脑出血后1d即有Ang-1表达,模型组至21d达到高峰,随后开始下降,益气组、活血组及益气活血组表达高峰时间提前至14d,活血组及益气活血组至21d仍维持一定的表达水平;各个时间点益气活血组表达均高于其他组。结论:益气活血中药可能通过使脑出血血肿周围脑组织Ang-1的表达增强,而促进脑出血损伤区血管新生和成熟,并促进脑出血大鼠神经功能的恢复。 Objective: To investigate the mechanisms of method of supplementing Qi and activating blood circulation by observing its effects on expressions of angiopoietin- 1(Ang-1) in brains of rats with intracerebral hemorrhage (ICH). Methods: ICH model was induced by injecting collagenase typeⅦ stereotaxically into right globus pallidus. One hundred and fifty-five SD rats were randomly divided into sixgroups, including normal control group (n=5), sham operation group, ICH model group, method of supplementing Qi and activating blood circulation-treated group, supplementing Qi-treated group and activating blood circulationtreated group (each n=30). The last treated 3 groups were administered orally with the classic method of supplementing Qi and activating blood circulation prescription of Buyang Huanwu decoction, its supplementing Qi and activating blood circulation components, respectively. The expressions of Ang-1 were assayed by Western Blotting on 1, 4, 7, 14, 21 and 28 days after the onset in 5 rats of each group. Results: Studies showed that the expressions of Ang-1 in the normal control and sham operation groups had no significant changes at different time points. The expressions of Ang-1 in the model and three medically treated groups were observed on 1 day, the expressions increased gradually, peaked on 21 day in model group and then decreased. The peak time of supplementing Qi-treated group, activating blood circulation-treated group and of supplementing Qi and activating blood circulation-treated group was advanced on 14 day, the expressions of supplementing Qi and activating blood circulation- treated group stayed on high levels to 28 day. Ang-1 expressions of supplementing Qi and activating blood circulation-treated group were significantly higher than other groups at every time points. Conclusion: The intervention of expressions of Ang-1 by method of supplementing Qi and activating blood circulation are up-regulated, which may play an important role in the process of angiogenesis following ICH, promoting and microvascular net work reconstruction and functional recovery of the rats' nerve following cerebral hemorrhage.
