肢体缺血再灌注后肝损伤中细胞凋亡的发生及牛磺酸的保护效应 被引量:2

Apoptosis in liver injury following limbs ischemia reperfusion and the protective effects of taurine

摘要 目的观察大鼠肢体缺血再灌注(LIR)后肝损伤和肝脏细胞凋亡情况及其发生机制,并探讨牛磺酸的保护效应。方法实验采用Wistar大鼠30只,建立大鼠LIR损伤模型,随机分为对照组、缺血再灌注(IR)组和牛磺酸+缺血再灌注(TR)组,每组10只。分别测定各组大鼠肝组织丙二醛(MDA)、黄嘌呤氧化酶(XOD)、髓过氧化物酶(MPO)及Ca^2+含量的改变;利用琼脂糖凝胶电泳观察DNA梯状条带,利用TUNEL法检测各组肝脏细胞的凋亡情况。HE染色观察肝脏形态学变化。结果与对照组比较,IR组大鼠肝组织MDA、XOD、MPO、Ca^2+含量均明.显升高(P均〈0.01),肝脏细胞DNA琼脂糖凝胶电泳可见梯状条带;TUNEL染色结果显示凋亡百分率明显升高(P〈0.01)。与IR组相比,TR组血浆和肝组织中MDA、XOD、Ca^2+含量和MPO反均有所下降(P〈0.01或〈0.05),肝脏细胞DNA琼脂糖凝胶电泳显示梯状条带不明显;凋亡百分率降低(P〈0.01)。结论细胞凋亡参与了大鼠LIR后肝损伤的发生,而牛磺酸可减轻大鼠LIR所致肝脏损伤及细胞凋亡的发生。 Objective To observe the apoptosis in liver injury following limbs ischemia-reperfusion(IR) in rats and the protective effects of taufine. Methods The model of limbs ischemia-reperfusion injury was established. 30 Wistar rats were randomized into 3 groups: control group, IR group and taurine + reperfusion group (TR group) ( n = 10 for each group). The levels of malondialdehyde (MDA) and xanthineoxidas (XOD) ,calcium and myeloperoxidase (MPO) in the liver tissue were measured. DNA fragmentation was observed and analyzed by agarose gel electrophoresis. Apoptosis was detected by TUNEL methods. The morphologic changes were observed with HE staining. Results Compared with control group, the values of MDA, XOD, MPO, calcium in liver tissue were increased significantly in IR group (P 〈0.01 ) ,but the values of those in TR group were lower than in IR group (P 〈0.01 ). The percentage of apoptosis cell was higher in IR group than in control group( P 〈 0.01 ). Compared with IR group, the percentage of apoptosis cell was lower in TR group ( P 〈 0.01 ). IR group presented DNA ladder pattern, while TR group showed no specific DNA ladder pattern in agarose gel electrophoresis. Conclusion Apoptosis participates in the liver injury after limb ischemia-reperfusion. Taurine can mitigate the liver injury and apoptosis after limb ischemia-reperfusion injury in rats.
