摘要
目的探讨大肠癌组织中基质金属蛋白酶-9(MMP-9)及其抑制物基质金属蛋白酶抑制剂-1(TIMP-1)的表达和临床意义。方法采用PowerVision二步法免疫组织化学染色检测42例大肠癌组织及其癌旁组织中MMP-9和TIMP-1表达情况并分析其与临床病理参数的关系。结果MMP-9在大肠癌组织中表达率明显高于癌旁组织(69.1%与45.2%,P=0.009);TIMP-1在大肠癌组织中表达率明显高于癌旁组织(61.9%与40.5%,P=0.004)。MMP-9的表达与Dukes分期、浸润深度、淋巴结转移正相关(rs=0.429,P=0.005;rs=0.372,P=0.015;rs=0.372,P=0.015);TIMP-1的表达与Dukes分期、浸润深度、淋巴结转移正相关(rs=0.394,P=0.010;rs=0.382,P=0.013;rs=0.382,P=0.013)。MMP-9的表达与TIMP-1的表达呈正相关(rs=0.641,P〈0.001)。结论MMP-9和TIMP-1在大肠癌发生发展中起重要作用,联合检测二者对大肠癌侵袭转移的预测、预后评估及指导合理的综合治疗等具有重要的临床意义。
Objective To investigate the expression and clinicopathological significance of matrix metalloproteinase(MMP-9) and matrix metalloproteinase tissue inhibiting factor-1 (TIMP-1) in eolorectal carcinomas. Methods Immunohistoehemistry (PowerVision method) in the paraffin embedded tissue samples was used to determine MMP-9 and TIMP-1 in 42 eolorectal carcinomas and its adjacent normal colorectal mueosa. The relationship of their expression with some clinicopathological characteristics was analyzed. Results MMP-9 and TIMP-1 were both significantly overexpressed in colorectal carcinomas compared with its adjacent normal colorectal mucosa (69.1% and 61.9% vs. 45.2% and 40.5% ,P = 0.009 and P = 0. 004), and MMP-9 was positively associated with the regional lymph node metastasis, bowel wall invasion and Dukes stage ( rs = 0. 372, P = 0.015 ; L = 0. 372, P = 0. 015 ; rs = 0.429 ,P = 0.005 ). TIMP-1 was positively associated with Dukes stage, bowel wall invasion and regional lymph node metastasis (rs =0.394 ,P =0.010;r, =0.382,P =0.013 ;rs = 0. 382,P =0.013 ). There was a positive correlation between the expression of MMP-9 and TIMP-1 ( rs = 0. 641, P 〈 0. 001 ). Conclusion MMP-9 and TIMP-1 may play a key role in eoloreetal carcinogenesis. Examination of MMP-9 and TIMP-1 expression may have an important significance for evaluating prognosis, predicting of invasion and metastasis and comprehensive therapy.
