叠鞘石斛茎段组织培养与花芽诱导 被引量:7

摘要 目的研究叠鞘石斛的快速繁殖及离体开花的培养体系。方法选用野生的叠鞘石斛带侧芽的茎段作为外植体,培养在附加不同激素的培养基上。结果MS培养基附加6-BA(1.0 mg.L-1),NAA(0.5 mg.L-1)对侧芽的诱导效果最佳。而MS培养基附加6-BA(2.0 mg.L-1),NAA(0.4 mg.L-1)最适合丛生芽的增殖。把无菌苗培养在附加6-BA(2.0 mg.L-1)的MS培养基中,少数植株被诱导开花,诱导率为10.0%。结论用带腋芽的茎段作为外植体在MS培养基中,附加一定的6-BA和NAA可以诱导丛生芽的产生。 Objective To study rapid propagation and in vitro flowering system for Dendrobium denndanum. Methods Stem with lateral buds of wild denndanum was used as explants and cultured on various culture media with different portions of hormone. Results The best medium for lateral buds induction was the MS with addition of 6 - BA ( 1.0 mg · L^- 1 ) , NAA (0.5mg/L) , while MS with the addition of 6 - BA ( 2.0mg · L^- 1 ), NAA ( 0.4mg · L^- 1 ) was suitable for the cluster shoots propagation. A few plantlets were induced flowering after cultured on MS medium with addition of 6 - BA (2.0 mg · L^- 1 ) , the frequency of floral bud induction was 10.0%. Conclusion The cluster shoots can be induced from the segments with lateral buds on the MS basic medium with addition of 6 - BA and NAA.
