超声联合微泡对体外骨髓干细胞影响的实验研究

摘要 目的研究不同距离超声辐射微泡对体外培养干细胞的影响,为超声微泡在干细胞移植领域的临床应用提供必要的实验依据。方法将体外培养的干细胞随机分为对照组和超声联合微泡实验组。观察不同距离超声辐射微泡对干细胞存活率、细胞凋亡以及细胞超微结构变化的影响。结果①实验组与对照组相比:存活率明显低于对照组,凋亡率明显高于对照组;电镜下对照组细胞超微结构无改变,实验组细胞结构有改变。②实验组内比较:随着超声辐射距离的增加,细胞的存活率逐渐增加,而细胞凋亡逐渐减少;细胞碎片逐渐消失,超微结构变化逐渐减少;可逆性微孔也随照射距离增加而逐渐增多。结论超声联合微泡造影剂在8cm以内对干细胞的存活率、凋亡率及细胞超微结构影响较大,在8cm以外对骨髓干细胞影响不明显,是安全的。 Objective Study the effect of ultrasound-mediated microbubble in different distance on MSCs cultured in vitro, in order to provide an evidence for clinical application of ultrasound-mediated microbubble in stem cell therapy. Methods MSCs of pigs are random divided into control group and experimental group; according to the distance from ultraphonic transducer to MSCs, is divided into group; ultrasound (1 MHz,2 W/cm^2), mireobubble (Sono Vue). Results are observed after MSCs transplanted 0 hour and after MSCs transplanted 24 hour through trypan blue staining, transmission electron microscope and FCM. Results It was confirmed that the ultrasound-mediated microbubble affect MSCs in forming microbore at exceed the scope of 8 cm, and the effect was not obvious in apoptosis and cytoactive. Conclusion It was safy that ultrusound-mediated microbubble on MSCs at exceed the scope of 8 cm.
