摘要
目的研究帕金森病(PD)黑质多巴胺神经元的死亡机制。方法用脱氧核糖核酸(DNA)标记法,借助荧光显微镜观察多巴胺(DA)和还原型谷胱甘肽(GSH)对PC12细胞凋亡的影响。结果发现DA可诱发PC12细胞凋亡,适中浓度时(0.45mmol/L)PC12细胞凋亡数最多(凋亡率为48.7%±6.3%);抗氧化剂还原型谷胱甘肽(GSH)可部分阻止DA诱发的PC12细胞凋亡(P<0.01)。结论凋亡参与了PD的病变过程,采用适当的抗氧化剂对于PD治疗具有积极的意义。
Objective To study the mechanism of dopaminergic neuronal death in substantia nigra of Parkinson,s disease(PD).Method The effect of dopamine(DA) and glutathione (GSH) on apoptotic PC12 cell were examined by fluorescence microscopy using the DNA endlabeling methods.Results It has been demonstrated that DA may induce apoptosis in PC12 cell,moderate concentrations of DA (0.45mmol/L) gave maximal number of apoptotic cell (the rate of apoptosis:48.7%±6.3%);and the antioxidant reduced glutathione(GSH) may partially prevent DAinduced apoptosis in PC12 cells (P<0.01).Conclusion Apoptosis may be involved in the pathogenesis of PD,and treatment with proper antioxidants may have positive influence on the therapeutic strategy for PD.
