摘要 目的研究帕金森病(PD)黑质多巴胺神经元的死亡机制。方法用脱氧核糖核酸(DNA)标记法,借助荧光显微镜观察多巴胺(DA)和还原型谷胱甘肽(GSH)对PC12细胞凋亡的影响。结果发现DA可诱发PC12细胞凋亡,适中浓度时(0.45mmol/L)PC12细胞凋亡数最多(凋亡率为48.7%±6.3%);抗氧化剂还原型谷胱甘肽(GSH)可部分阻止DA诱发的PC12细胞凋亡(P<0.01)。结论凋亡参与了PD的病变过程,采用适当的抗氧化剂对于PD治疗具有积极的意义。 Objective To study the mechanism of dopaminergic neuronal death in substantia nigra of Parkinson,s disease(PD).Method The effect of dopamine(DA) and glutathione (GSH) on apoptotic PC12  cell were examined by fluorescence microscopy using the DNA endlabeling methods.Results It has been demonstrated that DA may induce apoptosis in PC12 cell,moderate concentrations of DA (0.45mmol/L) gave maximal number of apoptotic cell (the rate of apoptosis:48.7%±6.3%);and the antioxidant reduced glutathione(GSH) may partially prevent DAinduced apoptosis in PC12 cells (P<0.01).Conclusion Apoptosis may be involved in the pathogenesis of PD,and treatment with proper antioxidants may have positive influence on the therapeutic strategy for PD.
