摘要
目的:建立快速测定血清山莨菪碱(anisodamine,Ani)含量的方法,并测定家兔经灌胃(ig)及静注(iv)Ani的药代动力学参数和绝对生物利用度(Fabs)。方法:反相HPLC与紫外检测器联用,以甲醇-水-三乙胺(30∶70∶0.15)为流动相,内标法测定兔血清中Ani浓度,并代入非线性拟合(NLFT·ZHU)程序提取有关参数。结果:兔灌胃及静注Ani后其药动学行为符合二室模型,属快分布型药物,Fabs为6.36%。结论:建立并优化了快速、高效、灵敏的反相HPLC检测Ani的方法。该方法线性范围宽,日内误差和日间误差分别平均为2.4%和2.7%,并基本阐明了Ani在兔体内的经时过程。
Objective: To build a rapid method for the separation and determination of anisodamine hydrochloride in rabbit's serum, and determining the pharmacokinetic parameters after iv or ig Ani and its absolute bioavailability in rabbit. Methods: Determining the serum concentration of Ani in rabbit by reversed phase HPLC and UV detector at 214 nm, CH 3OH H 2O (C 2H 5) 3N(30∶70∶0.125) as mobil phase, atropine sulfate as an internal standard. Calculation of the pharmacokinetic parameters by using a NLFT.ZHU program on microcomputer. Results: A two compartment open model featured the pharmacokinetic process after iv or ig Ani in rabbits, belong to fast distribution drug, absolute bioavailability is 6.36%. Conclusions: A rapid, sensitive and precise method for the separation and determination of Ani by RP HPLC has been developed and optimized. The pharmacokinetic process after iv or ig Ani in rabbits has been expounded.
