摘要
通过正交试验获得了磷脂酶A1催化水解大豆油的最佳水解条件:水解温度35℃、加酶量26U/g、加水量40%,水解时间8h。在此条件下,以KOH计,水解体系上层油层酸值达65.51mg/g。当用0.1%的氯化钙水溶液作为水解反应物时,和蒸馏水相比,钙离子对维持磷脂酶A1多次循环利用的残余活力有促进作用。水解反应残余活力的磷脂酶A1再加上50%的新添加酶,表现出很强的催化水解活力,酶回收使用6次时,残余酶活力仍然有初次使用活力的86.88%。
The optimal conditions for partial hydrolysis of soybean oil catalyzed by phospholipase A1 were obtained by the orthogonal experiment, which were reaction temperature 35℃, water content 40 wt%, enzyme dosage 26 U/g and reaction time 8 h. Under these conditions, the acid value of reaction mixture in the upper oil layer reached of 65.51 mg KOH/g. Using 0.1% of calcium chloride solution as the reactant at the circulated reactions, the residual activity of the enzyme was improved by calcium ion. The mixture of recycled enzyme and 50% of fresh phospholipase A1 showed high hydrolysis activity. After used for six batches, the residual enzyme activity reached 86.88%.
