大鼠血小板杀菌活性研究

摘要 目的:探讨血小板在宿主防御细菌感染中的作用。方法:Wistar大鼠麻醉后腹主动脉取血,离心分离出富血小板血浆(PRP),从PRP中分离血小板,以不同的诱导剂[凝血酶、二磷酸腺苷(ADP)和肾上腺素(AD)]分别诱导血小板及PRP,观察其杀菌活性。结果:分别经ADP、AD刺激的PRP和血小板均无杀菌作用,而血小板和PRP经凝血酶刺激后,均具有较强杀灭革兰阳性(G+)细菌的作用,但对革兰阴性(G-)细菌却无杀灭作用。结论:大鼠血小板经诱导的可释放杀菌物质,在机体防御细菌感染时发挥重要的作用。 Objective: To explore the role of rat platelet in host defense bacteria infection. Methods: Blood was collected from Wistar rats. The blood was centrifuged to prepare platelet-rich plasma(PRP), the platelets were then pelleted from plasma by centrifugation. The platelet and PRP were stimulated by several agents(thrombin, adenosine diphosphate and adrenaline hydrochlaride), and then the bactericidal activities were determined by counting bacterial colony formed unit(CFU) after incubation for 2h at 37℃ and the bactericidal rate calculated. Result: Platelet and PRP upon stirnulatied by adenosine diphosphate(ADP) and adrenalin(AD) did not show bactericidal. Platelet and PRP stimulatied with thrombin had potent bactericidal activity toward Gram-positive (G+) bacteria, but no effect on Gram-negative (G-)bacteria. Conclusion: Rat platelet with thrombin-induced can release antibacterial peptides and play an important role in host defense against bacteria infection.
