摘要
目的在以脱矿脱细胞骨基质环为支架以纤维环细胞为种子细胞构建组织工程化椎间盘纤维环细胞支架复合体过程中,探索最佳的构建复合体方法。方法分别使用纤维蛋白凝胶接种技术和传统的直接接种技术-静置法构建技术构建组织工程化椎间盘纤维环细胞支架复合体。对构建产物进行倒置显微镜观察、扫描电镜观察和细胞计数,比较两方法的效果。结果纤维蛋白凝胶接种技术构建的细胞支架复合体,细胞粘附更多、增殖更迅速。结论纤维蛋白凝胶接种技术在以脱矿脱细胞骨基质环为支架以纤维环细胞为种子细胞构建组织工程化椎间盘纤维环细胞支架复合体过程中,比静置法效果更好。
Objective To explore which method is better in construction of the cell-scaffold complex by anulus fibrosus seed ceils and demineralized and decellular scaffolds for intervertebral disc anulus fibrosus tissue engineering. Metheds The anulus fibrosus seed cells were planted into the demineralized and decellular scaffold with two difference methods,static precipitation method and fibrin gelatum inoculation method. The cell-scaffold complex was examined by inverted microscope,scanning electron microscope and cell counting. Results In the cell-scaffold complex constructed by fibrin gelatum inoculation method,cells could adhere more in quantity,distribute more uniform and proliferate more quickly. Conclusion The fibrin gelatum inoculation method is better in construction of the cell-scaffold complex by anulus fibrosus seed cells and demineralized and decellular scaffolds for intervertebral disc anulus fibrosus tissue engineering.
